American Gary Woodland shot a bogey-free round to lead Justin Rose by two shots at Pebble Beach.
Englishman Rose holed a one-under 70 to move to seven under, while American Woodland carded a 65, leapfrogging Rose going to nine under.
Rose started on the back nine, and extended his lead to three with birdies at 15 and 18.
The 38-year-old then bogeyed the first in a strengthening wind, but responded with a birdie at the second after a brilliant seven iron put him within a few feet of the pin.
He complained about a spectator's camera usage at four - where he bogeyed, before finishing with five pars for his first back-to-back below-par scores in 42 rounds at the US Open.
35-year-old Woodland from Kansas hit a 50-foot putt at the 18th for his sixth of the day to end an impressive display.
The world number 25 has won three PGA Tour titles since turning professional in 2007.
He now leads at a major after 36 holes for only the second time in his career - after the 2018 US PGA Championship, where he finished tied for sixth.
He has never converted a halfway lead into a tournament win in six attempts.
Rory McIlroy was two under for the day, and finished on ahead of Brooks Koepka, chasing his third successive US Open.
The Northern Irishman enjoyed a drama-free front nine with birdies at four and seven.
The four-time major champion was tied for second at six under at the 11th, before he slipped away.
A bogey at 13 and double bogey at 14 lost his momentum, the latter of which had a tricky pin position where the ball rolled back off the green before McIlroy chipped into the greenside bunker.
He recovered well with a nine iron onto the green at the 15th, setting up a birdie, befor a stunning putt on the fringe at the back of the 16th green.
Koepka started his round in 10th, and holed three birdies and a bogey to stay in contention, he's bidding for a fifth major title in his last nine appearances.
Americans Chez Reavie and Chesson Hadley join Koepka on four under, alongside Matt Kuchar who eagled at the 18th to go alongside his compatriots.
Louis Oosthuizen sits third after a one-under seventy, while Tiger Woods finished with back-to-back bogeys to end level.
Meanwhile compatriot Aaron Wise finished with two birdies for a 71, and sits level with McIlroy.
Having missed the cut at the last two US Opens, Matt Wallace improved with a 68 to go four under overall.
Welshman Rhys Enoch's round was bettered only by the leader, posting a 66 to make the cut at two over.
Rickie Fowler was five under after day one, but the 30-year-old American holed five bogeys and a double bogey in a second round of 77 to fall off the pace.
2010 US Open champion at Pebble Beach Graeme McDowell made five birdies in seven holes on the front nine, before bogeys at the 11th and 14th saw him also fall back.
Ian Poulter also made five birdies, but a quadruple-bogey eight at the third hole saw him miss the cut for the third time in his last five majors.
Englishmen Matt Fitzpatrick, Luke Donald, Paul Casey, Tommy Fleetwood and Tyrell Hatton all made the cut but Lee Slattery missed out by a stroke after carding a double bogey seven on his final hole.
