The Australian hit three more birdies on the back nine, before three putting at the par-three 17th.
Former champion Phil Mickelson was often in the rough, but finished level at the halfway point in the tournament.
New Zealand's Danny Lee, who finished the opening round second behind Koepka on six under carded a four-over 74 to slip back to two under.
