After a day which saw 18 wickets, England couldn't take a single one today.
Holder and Dowrich were not out for 202 and 116 respectively.
The former declared on 415-6 after surpassing 200, setting a target of 628 for the tourists to win.
Holder's double century included 23 fours and 8 sixes.
England openers Rory Burns and Keaton Jennings reached 56-0 at the close of play.
The best England can hope for is a draw from this first Test, but even this is wishful thinking, despite the Kensington Oval pitch having flattened out.
Instead, the West Indies need 10 wickets to take a 1-0 lead in the three-Test series.
